Overview

Mode of Action (MoA) inactivates Cascade by trapping the PAM-recognising Cas8 subunit in a non-productive conformation, incapable of performing PAM recognition.
Evidence DMS3m phage engineered to express acrIC8 was tested on PAO1IC and a Type I-E strain (PA4386). The phage regained full infectivity on both (EOP ≈ 1), and CRISPRi assays confirmed inhibition of DNA binding. AcrIC8 is a broad-spectrum Acr that blocks both Type I-C and Type I-E systems.
