Ectopic expression of AcrIIA24 conferred resistance to CRISPR targeting in E. coli expressing St3Cas9 (ECO_0000017). AcrIIA24 was cloned into a plasmid and co-expressed with St3Cas9 and a targeting sgRNA. In plasmid interference assays, AcrIIA24 significantly restored bacterial colony formation. In vitro cleavage assays showed that AcrIIA24 did not prevent DNA binding by Cas9 but strongly inhibited cleavage.
