Evidence Deletion of the genomic locus encoding AcrIIA25 from its native mobile element resulted in loss of inhibitory activity against SpyCas9 in E. coli (ECO_0007379). AcrIIA25 was cloned into an inducible expression plasmid and co-expressed with SpyCas9 targeting a plasmid. Plasmid interference assays showed moderate restoration of colony formation. In vitro assays showed AcrIIA25 could inhibit DNA cleavage but had minimal effect on DNA binding. Phage assays confirmed restored phage infectivity in strains expressing AcrIIA25, indicating moderate anti-CRISPR function via inhibition of the cleavage step.
