In E. coli, co-expression of AcrIIA34 with Cas9 allowed bacteria to grow, showing it blocked Cas9 from cutting the plasmid. Lab tests confirmed it does not block sgRNA loading, but does block Cas9 from binding DNA, as shown by DNA binding (EMSA) experiments (ECO_0001807).
