Deletion of ORF35 from ΦSMS22 (ECO_0001038) resulted in reduced infectivity against E. coli expressing the Tmn defense system. ORF35 (anti-Tmn) from ΦSMS22 was cloned into a plasmid and co-expressed with Tmn in E. coli. When challenged with phages, only ΦSMS22-derived anti-Tmn restored infectivity of phages that were otherwise blocked by Tmn (e.g., ΦKSS9), confirming its inhibitory activity against Tmn (ECO_0000017).
