Evidence
|
ArdR was shown to repress expression of both ardA and ardB, particularly the downstream P1 promoters, through deletion mapping and promoter activity assays using CUP-region constructs. Promoter probe plasmids (e.g., pCAT66-6) showed significant chloramphenicol acetyltransferase (CAT) activity in the absence of ArdR, which was abolished upon its expression, indicating ArdR-dependent repression. Maxicell analysis confirmed expression of a ~20-kDa protein from ardR. The gene is located adjacent to ardB, and deletion of ardR led to derepression of ardB-mediated antirestriction activity. These results demonstrate that ArdR acts as a transcriptional repressor of both ard genes and can function synergistically with ArdK to silence both P1 and P2 promoters.
